COLORTRIX TECHNICAL OVERVIEW

COLORTRIX has been designed specifically to be coupled to PATHATRIX as a rapid presumptive positive screening tool.

The COLORTRIX procedure involves adding an antibody/enzyme (Horse Radish Peroxidase - HRP) conjugate, known as "Reagent A" to the PATHATRIX concentrate for 5 minutes. The sample is then diluted with 1ml of wash buffer and the conjugate labelled bead/bacteria complexes are recovered using a magnet. After a further two washes half the concentrate is removed and added to "Reagent B" (which is a pre-prepared substrate for the HRP), and the sample is left for 5 minutes to develop colour.

A "Blue" colour is recorded as "Presumptive Positive". Absence of colour development is recorded as "Negative". If a "Presumptive Positive" is recorded the sample remaining in the wash vessel can then be plated onto the appropriate agar media or confirmed by molecular/biochemical techniques. If a "Negative" result has been obtained no further action is required.

There are two formats of the COLORTRIX assay:

  • Couples to the "30 minute" PATHATRIX format
  • Couples to the "same day" E.coli O157 & Salmonella PATHATRIX formats
