PATHATRIX is a rapid detection method for pathogens, spoilage bacterium and viruses in food, environmental and bio-threat samples. The PATHATRIX is a novel patented RIMS (re-circulating immuno-magnetic separation) technology that relies on the use of antibody coated paramagnetic particles to selectively bind and purify the target organism from a comprehensive range of complex matrices. By re-circulating the sample over a "capture phase," comprised of immobilized antibody coated magnetic beads, this increases the sensitivity of the capture. A high volume wash enables the efficient removal of the sample matrices, non-specific microorganisms and PCR inhibitors. Pathatrix consumables are available in two convenient formats:
Once captured and concentrated the beads are ready for use with a variety of detection methods: direct plating; PCR; ELISA etc. The Pathatrix consumable is loaded onto the instrument with the pre-enriched sample and is circulated for 30 minutes.
The beads are added immediately prior to re-circulation and the beads become immobilized on the capture phase.
The beads are added immediately prior to re-circulation and the beads become immobilized on the capture phase.
During re-circulation, at approx 500 ml per minute, the target microorganism is captured. The high flow rate significantly reduces non-specific binding.
After capture is completed, a high volume (50-100 ml) wash efficiently removes the sample matrices, non-specific miroorganisms, and PCR inhibitors.
Following the wash the removal of the magnet enables the bead/target complexes to be released and concentrated ready for detection. DETECTION SOLUTIONS
HOW FAST DO YOU WANT RESULTS? The time to result is dictated by the pre-enrichment incubation of the sample (4-24 hours) and the speed of the end point detection used (40 mins - 48 hours). Pathatrix enables results to be available in as little as 5-6 hours from receipt of sample, but this can be tailored to suit the customers' individual needs. AOAC-RI APPROVED? Data from internal and external validation studies e.g. AOAC-RI trials have demonstrated that the PATHATRIX system is significantly more sensitive than many of the current standard methods, at low spike levels 1-10 cfu/25g sample. Full AOAC RI validation reports are available on request together with an extensive bibliography of published data (Technical bulletin).
